چکیده

In this work, a xylanase-producing strain, Streptomyces griseorubens LH-3, was cultured, and the crude xylanase was prepared. Analysis of its enzymatic properties revealed that the crude xylanase possessed good thermal stability at temperatures below 60 o C, exhibited a wide pH range from 4.0 to 9.0, and was cellulase-free. This crude enzyme was used to treat eucalyptus kraft pulp, and the release of chromophores was the highest at the dosage of 20 IU g −1 dry pulp. Compared with the untreated group, biobleaching of eucalyptus kraft pulp with this enzyme increased the brightness of the pulp by 12.9% and reduced the Kappa number by 27.4%. Biobleaching of eucalyptus kraft pulp with this enzyme obtained the same final pulp brightness compared with that of the control; however, hydrogen peroxide consumption was reduced by 17% and the yield and viscosity of the pulp was increased by 1.47% and 1.53%, respectively. This crude xylanase has promising potential for industrial applications.

Evaluation of a New Laccase Produced by Streptomyces Ipomoea on Biobleaching and Ageing of Kraft Pulps

The aim of this work is to prove the suitability of a new alkaline and halotolerant bacterial laccase (SilA) produced by Streptomyces ipomoea CECT 3341 to enhance the conventional chemical bleaching process of an industrial eucalyptus kraft pulp. The laccase used for this study was a recombinant laccase obtained from cultures of E. coli BL21 (DE3) grown in LB liquid medium. The biobleaching exp...

Kraft pulp biobleaching and mediated oxidation of a nonphenolic substrate by laccase from Streptomyces cyaneus CECT 3335.

A new laccase (EC 1.10.3.2) produced by Streptomyces cyaneus CECT 3335 in liquid media containing soya flour (20 g per liter) was purified to homogeneity. The physicochemical, catalytic, and spectral characteristics of this enzyme, as well as its suitability for biobleaching of eucalyptus kraft pulps, were assessed. The purified laccase had a molecular mass of 75 kDa and an isoelectric point of...

Biobleaching application of cellulase poor and alkali stable xylanase from Bacillus pumilus SV-85S

The potential of extracellular alkali stable and thermo tolerant xylanase produced by Bacilluspumilus SV-85S through solid state fermentation was investigated in pulp bleaching in association with conventional bleaching using chlorine and chlorine dioxide. The biobleaching of kraft pulp with xylanase was the most effective at an enzyme dose of 10 IU/g oven dried pulp, pH 9.0 and 120 min incubat...

Enhanced Production of Cellulase-free Xylanase by Alkalophilic Bacillus Subtilis Ash and Its Application in Biobleaching of Kraft Pulp

This paper reports high level production of a cellulase-free xylanase using wheat bran, a cost-effective substrate, under submerged fermentation by alkalophilic Bacillus subtilis ASH. Production of xylanase was observed even at alkaline pH up to 11.0 and temperature 60 °C, although the highest enzyme titer was recorded at neutral pH and 37 °C. The enzyme production under optimized fermentation ...
